Alzheimer's Disease: Tau Pathology and Dysfunction of Endocytosis

Alzheimer’s disease (AD) is the most common form of dementia. Its prevalence will significantlyincrease in the coming decades, whereas no efficient treatment is currently available. AD hastwo main neuropathological lesions: amyloid plaques and neurofibrillary tangles (NFTs). Amyloidplaques are composed of amyloid s (As) peptides cleaved from the Amyloid Precursor Protein(APP) (Glenner and Wong, 1984). NFTs, present in the brain of AD and related neurodegenerativediseases, are constituted of tau proteins (Brion et al., 1985) in hyperphosphorylated and aggregatedform (Wang and Mandelkow, 2016).
