Generation of three‐dimensional pharmacophore models

Different approaches to the generation of pharmacophore models—pharmaco-phore discovery—are compared, and analyzed in detail at the level of their conceptual basis, their theoretical foundations, and the algorithms used to implement them. Strengths and weaknesses of each are discussed, with the aim of pointing toward future developments in this field.
