Methods for plasmid purification had been designed in the past predominantly to be easy to use in cloning and mutagenesis experiments, in which µg quantities were sufficient and quantitative yield was not an important issue. Current concepts in gene therapy involving plasmid DNA as vector have created a growing market for production scale quantities (1 g → 1 kg) of supercoiled DNA of clinical quality.
